Description of the Service Centre
The laboratory of FTICR-MS and Structural Mass Spectrometry provides the highest performance technology for problem solving in health and life sciences. Provides support to food and wine industry, forensics and fundamental research in chemistry, biochemistry and biology. It is the main laboratory of the Portuguese Mass Spectrometry Network, a National Roadmap Research Infrastructure of Strategic Interest.

Preserving non-covalent interactions in the gas-phase allows the analysis of intact protein complexes and macromolecular assemblies. This includes multimeric proteins, ligand-protein binding interaction studies, structure and stability analysis of proteins with quaternary structure. Bioconjugate analysis. Supramolecular chemistry applications and nanoparticle analysis, provided these are amenable to ionisation. Examples: Enzyme substrate interactions, dimeric and higher order protein structures, characterization of monoclonal antibodies (mAb).

Exact mass can be determined, within 1 ppm for just about any molecule. Most commonly used for molecular formula confirmation or in assisting in the determination of molecular formulas. Can be performed with pure compound or in complex mixtures, without chromatographic separation. Examples: Chemical products used in agro, pharma and food industries. Organic synthesis. Identification of contaminants.

Identification of metabolites based on ultra-high resolution mass spectra from cell or tissues extracts.Includes all classes of metabolites that may be extracted and ae amenable to ionisations. Lipidomics is a sub-set of metabolomics. Examples: Yeast, leishmania and other microorganism, animal cells in culture. Plant and animal tissue extracts.

Characterization of intact proteins, either pure or in mixtures. In the case of protein mixtures, chromatographic or electrophoretic separation is mandatory previous to mass spectrometric analysis. Accurate mass determination, with isotopic resolution. MSMS by CID or ECD to obtain sequence tags or protein identification without trypsin hydrolysis. Examples: Identification and characterization of post-translational modifications, protein isoforms. protein identification without enzymatic proteolysis.

The highest resolving power and most accurate mass spectrometer currently available in Portugal. It provides ultra-high resolution for the characterization of complex samples (crude oil, metabolomics, intact top-down protein analysis). It is equipped with the dual ionisation source Apollo II and can used in electrospray (ESI), nanoelectrospray (nanoESI) and matrix assisted laser desorption ionisation (MALDI). The mass spectrometry has MSn ability (n equal or greater then 2) with several physical fragmentation techniquessuch as CID, ECD, IRMPD and SORI-CID. Can be coupled to a capLC (Agilent 1100) or a nanpUHPLC (Ultimate 3000 RSLC).

Q-TOF Ultima from Micromass modified by MSVision for native mass spectrometry.The modifications include an extended m/z range, increased collision energy on the collision cell and increased range for ion isolation in the quadrupolo. Ionisation is achieved by nano electrospray (nanoESI). The system is capable of MSMS by CID using Ar or Xe.
